Description & Mechanism

FOXO4-DRI is a synthetic, modified peptide derived from regions of the Forkhead box O4 (FOXO4) transcription factor. It was engineered using a D-retro-inverso design to improve stability and resistance to proteolytic degradation in experimental systems.

In biochemical and cellular research models, FOXO4-DRI has been used to study protein–protein interactions between FOXO4 and p53. The peptide acts as a competitive inhibitor of this interaction, enabling investigation of transcriptional regulation, cellular senescence pathways, and stress-response signalling. Observed effects are dependent on experimental design, concentration, and model system, and are used for mechanistic exploration rather than defined biological outcomes.
