Description & Mechanism
5-Amino-1MQ is a synthetic methylquinolinium derivative designed as a potent inhibitor of nicotinamide N-methyltransferase (NNMT). In biochemical research systems, it modulates metabolic signaling by interfering with NNMT catalytic activity, thereby influencing levels of methylated nicotinamide derivatives and related metabolites. Peptide Sciences+1
NNMT is a cytosolic enzyme that catalyzes methyl transfer from S-adenosyl methionine (SAM) to nicotinamide and related substrates. Because of its metabolic role, NNMT is expressed in liver, adipose tissue, kidney, and other tissues. Peptide Sciences
In preclinical biochemical settings, inhibition of NNMT by compounds like 5-Amino-1MQ leads to alterations in intracellular NAD⁺ salvage pathways, shifts in methyl donor balance, and downstream metabolic signaling changes. Peptide Sciences
Applications in Research
- As a molecular probe to study NNMT function, regulation, and downstream metabolic pathways
- To investigate metabolic flux, methylation dynamics, NAD⁺ salvage, and related bioenergetic phenomena
- In cellular assays, to test effects of NNMT inhibition on gene expression, metabolite profiling, or metabolic phenotypes
- In animal / ex vivo experiments for mechanistic elucidation of metabolic regulation (e.g. adipose tissue, liver)
Handling, Reconstitution & Stability
- Weigh under inert/dry conditions; avoid moisture uptake
- For stock solutions, dissolve in water or dilute acid (e.g. 0.1 N HCl) or compatible solvent (e.g. DMSO) depending on assay needs
- Filter sterilize if needed (e.g. 0.22 µm) immediately prior to use
- Aliquot and store stock solutions at –20 °C (or lower) to avoid repeated freeze–thaw
- Monitor for precipitation or pH shifts before use in sensitive assays

Precautions & Notes
- Because 5-Amino-1MQ is a small molecule inhibitor with ionic character, buffer composition, ionic strength, and pH in assays may influence its behavior (ion pairing, binding).
- It is advisable to run appropriate controls (vehicle, dose–response) to exclude non-specific effects in in vitro systems.
- For in vivo or ex vivo studies (if employed in nonclinical contexts), consider potential biodistribution, clearance, and off-target interactions in experimental design.
- Use of light, high temperature, or prolonged aqueous exposure may degrade or oxidize the compound; prepare fresh or store stocks under protective conditions.
